CHLOROPHYL ANALYSIS
Extraction
- Remove samples from –70 freezer
- Record sample numbers on sample log sheet. Do not loose this sheet as it is the only record of the analysis we have. The volume filtered is 200ml and volume extracted is 10ml, unless otherwise noted
- Unfold samples and place in centerfuge vials. Do not use polystyrene vials Make sure surface of filter is completely exposed
- Add 10ml of 90% Acetone to vials
- Place sample vials in freezer and let extract for 24-48 hours
Analysis
- Turn on fluorometer (red button). Let warm up for approximately ½ hour
- Remove samples from freezer
- Shake samples and then centerfuge for 3 minutes
- Place 10ml of 90% Acetone in a borosilicate test tube. This is your blank sample
- Run a blank
- Press ENT key
- Press 2 for calibration
- Press 1 for blanking
- Press 1 for running a blank
- Place blank sample in fluorometer
- Once reading is stable press 0
- Wait 15 seconds for unit to complete reading. Press ESC. Press HOME
- Remove blank from fluorometer. You are know ready to run samples
- Decant sample into a borosilicate test tube
- Place test tube containing the sample into the fluorometer
- Once the reading has stabilized, record the value on the log sheet under Rb. Be sure to also note the sensitivity range
- Add 3 drops of 10% HCl to the test tube vial
- Once the value has stabilized, record the value on the log sheet under Ra. Be sure to also note the sensitivity range
- Once all samples have been analyzed and all values are recorded, dispose of samples in the waste bottle and test tubes in the glass disposal box
- Be sure to take a reading on the blank sample at the end of each run. Note any drift from zero
- Once all samples have been ran, turn off the fluormeter and wash the centerfuge vials with detergent. Triple rinse with deionized water after the detergent wash
